The generation of neuronal models in vitro has been a topic of sustained research, aiming to develop experimental setups to support mechanistic analyses and pave the way for the development of novel therapeutic approaches. A particularly challenging area is the development of advanced models suitable to emulate the human disease states. Currently, there is a critical need to develop in vitro models that resemble the in vivo environment as closely as possible. Among these models, organotypic culture systems have emerged as an important paradigm in neuroscience research. In the brain, cell types are highly organized into functional units, in which neurons and glia are tightly packed in an environment that is optimized to support their function. Attempts to reproduce this in vitro would greatly aid in the understanding of the mechanisms behind the maintenance of brain function, as well as the disruption of it. Here, we describe one of the most sophisticated, recently established in vitro paradigms – organotypic hippocampal culture. An important advantage of this approach is that tissue is maintained intact, which enables us to study both normal brain and pathological conditions in one model. We further discuss the advantages and disadvantages of this system, and how to optimally conduct experiments with this model. Moreover, we provide an overview of recent developments, which have expanded our knowledge of brain function and presented novel insights into the pathogenesis of neurodegenerative diseases, particularly Alzheimer’s disease.Crossroads Reunion Festival to Take Place August 11
